Time passes quickly when we are doing something we like, but when we stop and look back we realize that it has been two years since we published our latest Scientific Report and important changes and challenges have occurred since then.

In July 2015, the University of the Basque Country (UPV/EHU) and the CSIC entrusted me with the Direction of the Institute. First of all, I would like to recognize the excellent work carried out by the former Director, Prof. Félix M. Goñi.

At that time, one of our most immediate challenges was to move to a new building located in the Scientific Park – Campus Leioa, UPV/EHU. This long-awaited goal was finally materialized in February 2016 thanks to the strong and invaluable support from UPV/EHU and CSIC, as well as to the hard work and illusion of all members of our Institute. I strongly believe that people are the greatest value of our Institute, people with knowledge, diligence and passion in the pursuit of professional excellence. Finally, we thank the support and financial assistance provided by the Basque Government and the Biscay County Council, without which this would not have been possible.

At the end of 2015, in accordance with a spirit of continuous improvement, we were evaluated by a group of external experts of recognized prestige, whose recommendations helped us to better organize our research teams and optimize our resources. I would like to take this opportunity to thank all the members of this external committee for their outstanding work and, above all, their support, encouragement and involvement in improving our Institute.

On July 21, 2016, the Scientific Park was officially inaugurated and we had the opportunity to show our facilities and present some of our exciting research projects to political and scientific leaders, during a visit that I remember with pride and gratitude.

We have also changed our name, now “Instituto Biofisika”, in order to better reflect our growing status as a leading research institute, whose mission is to carry out basic and applied research in the field of biophysics and its applications to biotechnology and human health.

Now, we must continue working with determination and enthusiasm in order to conduct rigorous research, effectively disseminate and transfer the findings of this research, train the future generation of scientists in the field of biophysics and its applications, and, finally, serve society in as many ways as we can.

The Instituto Biofisika is a joint centre of the Spanish National Research Centre (CSIC) and the University of the Basque Country

(UPV/EHU).

The following pages contain a summary of our research activity during years 2015 and 2016. This report intends to serve a two-fold purpose, providing the reader with information about our activities, and supplying our sponsors with the necessary data about the end-product of their funding. “Euro-BioImaging appointed its Hub and nominates the fi rst generation of Nodes in December 2015”

One of those Nodes is the Advanced Light Microscopy Node Bilbao, Spain. The Bilbao Advanced Light Microscopy Node Candidate is a joint initiative of the imaging platforms of the Instituto Biofi sika and Achucarro Basque Center for Neuroscience, both located in the Science Park of the University of the Basque Country (UPV/EHU). The Node Candidate is focused on quantitative imaging of cellular events such as molecular interactions and biophysical studies such as lipid-mediated protein interactions or lipid-order imaging of model membranes and cells; and on the neurobiology of neuron-glia interactions.

The human immunodeficiency virus, or HIV, relies on two glycoproteins, gp41 and gp120, that form a non-covalent complex as the viral envelope. However, structural details of a transmembrane anchor found in gp41 were not known. In this paper high-resolution nuclear magnetic resonance spectroscopy was carried out to analyze this anchor. It was discovered that the anchor contains two helices that are held together by a flexible region. Further biophysical analyses revealed that the N-terminal helix was a scaffold for anti-MPER neutralizing antibodies. The Cterminal helix acted as an immunosuppressor and may contribute to the late stages of the viral fusion process into the host cell. It was concluded, “These data may guide the rational design of MPER vaccines and inhibitors”.

Carbon Nanotubes (CNTs) of lengths between 10 and 20 nanometers are inserted spontaneously into the plasma cell membrane. They present ionic transport properties similar to those of the cellular ion channels, which together with their non-protein biocompatible origin suggest them as candidates for the control of cell membrane excitability. We describe the protocol for obtaining CNTs for their study in cellular and artificial membranes.

This new research work, published in Nature Neuroscience reports that in Alzheimer’s disease, synaptic plasticity is altered by a protein originally described as a tumor suppressor: PTEN. In 2010, the research group of Dr. Esteban discovered that PTEN is recruited to synapses during normal (physiological) synaptic plasticity. This new investigation by Drs. Knafo and Esteban now indicates that this mechanism runs uncontrolled during Alzheimer’s disease. One of the pathological agents of the disease, the beta-amyloid, drives PTEN into synapses excessively, unbalancing the mechanisms for synaptic plasticity and impairing memory formation.

Although this is basic research using animal models, these studies contribute to dissect the mechanisms that control our cognitive function, and orient us towards potential therapeutic avenues for mental diseases where these mechanisms are deficient.

The phospholipid cardiolipin (CL) has been proposed to play a role in selective mitochondrial autophagy, or mitophagy. CL externalization to the outer mitochondrial membrane would act as a signal for the human Atg8 orthologs. The latter would mediate both mitochondrial recognition and autophagosome formation, ultimately leading to removal of damaged mitochondria. We have found that LC3B interacts preferentially with CL over other di-anionic lipids, that CL-LC3B binding occurs with positive cooperativity, and that the CL-LC3B interaction relies only partially on electrostatic forces. “Instituto Biofisika moves to a new building”

The Instituto Biofisika (CSIC, UPV/EHU) moved to the new facilities located in the Science Park of the Campus of Bizkaia. Since late February 2016, research groups and all the scientific instruments were operational in the new building. This new stage coincided with the change of name of this research center, which now is Instituto Biofisika instead of Biophysics Unit.

The new facilities and the new scientific program endorsed by an external committee and supported by the solid base established in the previous stage will allow facing the new challenges of society.
