• GEMFIN n=190; Response n=110.
• Covariates selected for calculating propensity scores (PS) included age, sex, history of thrombosis at time of resistance/intolerance, cytopenia at the lowest HU dose and JAK2 mutational status.
• Matched cohorts formed after PSM (n=92 each for ruxolitinib and BAT) were balanced for all covariates included in calculating PS

RESPONSE-2 Study Design
• Ruxolitinib-randomized patients had their doses individually titrated for efficacy and safety (to a maximum of 25 mg bid)
• Investigator-selected BAT as monotherapy included HU (at a tolerated dose if the patient were likely to receive benefit), interferon (IFN)/peg-IFN, anagrelide, pipobroman, immunomodulatory drugs, or no medication
• All patients received low-dose aspirin unless medically contraindicated

Conclusiones
• La elección del tratamiento de segunda línea tiene que adaptarse
a las características del paciente
• La hidroxiurea es la opción habitual en pacientes que se han
tratado en primera línea con interferón si bien no hay estudios que
hayan evaluado este aspecto
• El papel del interferón en segunda línea después de hidroxiurea
no está bien definido
• El busulfán requiere experiencia en el manejo y una adecuada
selección del paciente
• Ruxolitinib constituye una nueva alternativa para pacientes con
respuesta inadecuada o intolerancia a hidroxiurea con un
adecuado perfil de eficacia y seguridad
