Cyclodextrins in aqueous
solutions welcome “guest
molecules”, other than water, in
their hydrophobic cavities (Szejtli,
1998). The interaction of a CD
and a guest molecule has usually a
molar ratio of 1:1, but other kinds
of interactions can be found, as
showed in figure 3 (Stella et al.
2008). This complex formation
can then increase the solubility of
some drug molecules (Stella et al.
2008 and Loftsson et al. 1996).
Throughout the formation of
complexes between cyclodextrins
and drug molecules, no breakages
of covalent bonds are noticed, but
what leads to the formation of
such complexes is the release of
water molecules from the cavity
of the cyclodextrins,
hydrophobic interactions,
hydrogen bonding, among others (Loftsson 2004). Complexes formed by HP-0.6-β-CD and β-CD
with some bile salts: glycocholate (GC), glycodeoxycholate (GDC) and glycochenodeoxycholate
(GCDC) are the most relevant complexes for this report.

4.1.1 Binding Constant – K
Graphs indicating the K value for temperature 25, 30, 40 and 50ºC are shown below. All
temperature points show a decrease in K value, shifting the complexation equilibrium favoring
the reactants as temperature increases. It seems that GC has the highest decrease in K compared
to GDC and GCDC, where GC has decreased by 50%, GDC 30% and GCDC 32% from 25ºC to
50ºC. The CIs are smallest for GCDC and are largest for GC. This may be due to GCDC binding
more strongly with the CD, which gives a better “S” shaped curve when the ITC integrates the
peaks (observed in figure 8c). This can give a more accurate “one set of sites” iteration of the
data points.

stoichiometry of reaction, if the initial concentrations of the reactants are known (Ladbury and
Chowdhry, 1996). Heat absorbed or released after each injection is proportional to the change of
concentration of a bound ligand, described in the following equation:

For a reaction with one type of binding site for the molecule – M + nL ↔ MLn (for one type of
binding site n=1) and the cumulative heat can be calculated as follows:

Data plotting and fitting is obtained by plotting the 1st derivative of Q with respect to [L]total
against the molar ratio [L]total/[M]total (Holdgate, 2001). As equation shows, Q is dependent on Ka,
ΔH° and N. Those parameters can be obtained by fitting a line to data points (Ladbury and
Chowdhry, 1996).
Further on, when ΔH° and Ka values are known, the magnitudes of ΔG° and ΔS° can be
calculated from the following relationship:

Along with ΔG°, ΔH° and ΔS°, it is also possible to measure ΔCp – the change of heat capacity or
specific heat. ΔCp is used to predict the change of the other three parameters according to
different temperatures. The relationship is as follows:

The modification of some of the bands representing the guest molecule is indicative that only part of the molecule has been encapsulated by CD. The portion that has not been complexed is responsible for the presence of any unchanged bands

However, it is important to emphasize that when complexation occurs, the bands representing CD remain unchanged, and if the guest molecule does not present a very characteristic band, the changes may be imperceptible
1. Some authors report difficulty in
interpreting results due to the lack of a band representing the guest molecule that produces noticeable changes
40,57,58. There are still other authors who have
obtained inconclusive results59,60
.
Recent studies employing IR to evaluate the formation of inclusion complexes are presented in Table 2.
NUCLEAR MAGNETIC RESONANCE
Nuclear magnetic resonance (NMR) is a technique that has been widely employed to determine the formation of inclusion complexes, because it has the advantage of being able to reveal the structure of the complex by identifying the part of the host molecule that is included in the CD cavity

There are six protons in CD that can be used in the analysis of the NMR spectra of hydrogen (HNMR): three located in the outer surface (H1, H2 and H4), two in the cavity (H3 and H5), H3 near the wider exit and H5 near the narrow side. The last proton, H6, is closer to the narrow output of the cavity. The changes that occur in CD are usually in H3 and H5 and sometimes in H6, depending on the depth that the guest molecule enters the CD cavity
76,77, as illustrated in Figure 3.

Due to the formation of chemical bonds, when complexation occurs, signal changes take place in CD and guest molecule spectra60,78.
Another technique employed in the characterization of CD complexes is carbon 13 NMR (C13NMR). The results reveal the part of the guest molecule that has been encapsulated by CD, complementing the HRMN information
79.
Recent studies that use NMR to assess the formation of inclusion complexes are presented in Table 3.
CONCLUSION
A complete characterization of drug-cyclodextrin inclusion complex can be obtained using several analytical techniques as described in a previous review - part I. The analytical tools described in this review can show important details of the molecules whose, in addition with other techniques as phase solubility diagrams, dissolution studies and scanning electron microscopy can generate a more complete characterization of the formed complexes.

JUSTIFICACIÓN
Es relevante acomplejar al ácido ferúlico con ciclodextrinas ya que le brindará mayor solubilidad en soluciones acuosas y mayor estabilidad en el transcurso de su almacenamiento.
Los parámetros termodinámicos obtenidos por las técnicas de ITC y de espectroscopía de fluorescencia permitirán establecerán las fuerzas intermoleculares que gobiernan el proceso de inclusión del ácido ferúlico dentro de las ciclodextrinas.
Por otro lado el tener la estructura del acomplejamiento proporcionará información acerca de cómo esta unido estructuralmente el ácido ferúlico en las ciclodextrinas.

HIPÓTESIS
El ácido ferúlico presenta una estructura molecular
que permitirá la inclusión con determinada facilidad y
con asosiaciones específicas en cada una de las
ciclodextrinas (α, β, γ). Así mismo los métodos de ITC,
fluorescencia, IR y RMN permitirán obtener
información de la termodinámica y de la estructura del
proceso de acomplejamiento del ácido ferúlico con las
ciclodextrinas (α, β, γ).

Objetivo General
Evaluar los parametros termodinamicos del
proceso de encapsulamiento del acido ferulico
con ciclodextrinas, y la elucidacion estructural
de los complejos.

1. Realizar el analisis termodinamico del proceso de
acomplejamiento entre la ciclodextrina y el acido ferulico
utilizando la tecnica de calorimetria de titulacion isotermica
2. Obtener los parametros termodinamicos mediante el
ajuste de la curva de titulacion: la constante de union (Ku),
la entalpia (ΔHuo) y la estequiometria de union (n).

5. Elucidar la estructura la estructura de los complejos
del acido ferulico con las diferentes ciclodextrinas,
mediante resonancia magnetica nuclear (RMN)
unidimensional y bidimensional homonuclear.

1. Determinar los parametros termodinamicos
propios del proceso de acomplejamiento del acido
ferulico con las ciclodextrinas (α, β, γ) mediante
calorimetria de titulacion isotermica (ITC).
2. Determinar la contante de union (Ku) mediante
una titulacion fluorimetrica para contar con una medicion
alternativa.
3. Obtener la estructura de los complejos del acido
ferulico con las diferentes ciclodextrinas.

1.-Determinar el disolvente a emplear
Metanol (50 y 100%), Etanol (60, 70 y 100%),
NaOH (5 y 10%) Amortiguador de fosfatos 100 mM en un intervalo
de pH de 8 a 12
2.- Generar los espectros de IR
Mezclando 1mM de ácido ferúlico con las
diferentes ciclodextrinas 1 mM,
usando como disolvente metanol al
100%.
3.- Obtener los diagramas de fase-
solubilidad
Mediante la técnica descrita por Higuchi y
Connors del ácido ferúlico con la
ciclodextrinas al 100%.
4.- Realizar el analisis termodinamico del
proceso de acomplejamiento
utilizando la tecnica de calorimetria de
titulacion isotermica

1. Se logró disolver tanto el ácido ferúlico (3.0
mM) como a las ciclodextrinas (1 mM),
utilizando amortiguador de fosfatos 100
mM, pH 9.0, con agitación constante
durante 24 h a 30°C.
2. Se obtuvieron los espectros de IR para los
polvos del ácido ferúlico, cada una de las
ciclodextrinas así como para los
complejos.

La hidratacion de las CD’s es inebitable, los problemas que
pueda causar esto es:
Introducir variaciones en la medición de los parámetros
termodinámicos utilizando el calorímetro al incroporar màs agua
al medio de reacción.
El calor absorbido o liberado después de cada inyección es
proporcional a la variación de la concentración de un ligando
unido

CONDICIONES DE LA
EXPERIMENTACIÓN CON ITC
Acido ferulico 1X10-3 M
Ciclodextrinas (α-CD, β-CD, γ-CD) 1X10-2 M
Las condiciones de los experimentos seran:
20 inyecciones de 1.5 μl de solucio n de ciclodextrina en la celda
de muestra que contiene la solucio n de a cido ferulico.
El intervalo de tiempo entre las inyecciones sera de 300
segundos.
La velocidad de agitacio n sera de 310 rpm
La temperatura de la celda sera de 25 ºC
