El tamizaje de las librerias de compuestos
químicos incluye dos pasos fundamentales:
a) Expresión del Oct4-GFP y morfología celular
b) Estudios confirmatorios para tratar de dilucidar
el modo de acción del compuesto.
De unas 50 000 moléculas tamizadas poco más de 20 han mostrado tener la
actividad biológica deseada.
El entendimiento de los fenómenos biológicos a menudo se inicia
observándolos para luego diseñar o descubrir vías para perturbarlos.

CONTROL DE LA PRODUCCION DE TUMORES
We found that quercetin-induced selective cell death is caused bymitochondrial accumulation of p53 and is sufficient to prevent teratomaformation after transplantation of hESC- or hiPSC-derived cells. Takentogether, these results provide the “proof of concept” that small-moleculetargeting of hPSC-specific antiapoptotic pathway(s) is a viable strategy toprevent tumor formation by selectively eliminating remainingundifferentiated pluripotent cells for safe hPSC-based therapy.
CONTROL DE LA PRODUCCION DE TUMORES
Follow-up in vivo studies revealed that FL118 exhibits superior antitumorefficacy in human tumor xenograft models in comparison with irinotecan,topotecan, doxorubicin, 5-FU, gemcitabine, docetaxel, oxaliplatin, cytoxanand cisplatin, and a majority of mice treated with FL118 showed tumorregression with a weekly64 schedule. FL118 induced favorable body-weight-loss profiles (temporary and reversible) and was able to eliminatelarge tumors. Together, the molecular targeting features of FL118 plus itssuperior antitumor activity warrant its further development toward clinicaltrials.
